![]() ![]() ![]() The “south” in the name refers to the use of DNA probes, while the “west” refers to the protein blot. *end of open access rant*īowen and colleagues continued the naming convention in 1981 with their publication of the southwestern blot, a technique for identifying DNA-binding proteins in nuclear protein extracts using specific oligonucleotide probes. Ironically, considering the people power that was doubtless (at least partly) responsible for it’s eventual publication, Burnette’s paper is available only to Analytical Biochemistry subscribers. I began receiving phone calls from researchers unable to read the umpteenth photocopied generation of the pre-print, a sort of technical samizdat that I had to endlessly interpret”Ī few years later, Burnette eventually coaxed Analytical Biochemistry into accepting his paper and it was published in 1981 (see here), but by then, word of mouth had already beaten them to it. “…the few preprints I had sent to colleagues seemed to have undergone logarithmic Xerox multiplication. It happened because researchers, besides being interested in the technique itself, were tickled enough by its quirky name to make copies and send it to their friends. It happened through the sense of humor of the researchers who were doing the work, through people power (assisted by Xerox power). The reviewers hated it, they hated the name even more – obviously humor was not high on their agenda – and the manuscript was rejected.īut despite this, the popularisation of Burnette’s technique, and particularly the name “western blot” still happened even without the assistance of the literary establishment. Unfortunately for Burnette no sooner had he perfected his technique than a paper describing a very similar method, also inspired by nothern blotting, was published by Towbin et al working at the Friedrich Miescher Institute in Switzerland (see here for the reference and here for Towbin’s account of events).īurnette was dejected, but nonetheless, convinced that his methodology was sufficiently different to Towbin’s, he decided to submit a manuscript on his western blot method to the Analytical Biochemistry journal. Try pulling your boss up on that one next time he is in mid-flow talking about a “Northern blot” in a departmental presentation.Īnyway, back to our story. Note that among these techniques, only the Southern blot should be capitalised since it refers to Southern’s name, the others – nothern, western etc – are not proper nouns, so should not be capitalised. So if the Nowinski lab had been in New York, we would all be doing “eastern” blots.Ī quick aside for the pedants among us. Like nothern blotting, “western blot” was also an allusion to the Southern and nothern techniques, but Burnette had upped the ante by throwing in a geographical reference to location of the Nowinski lab. In a historic, but mostly forgotten conversation with Nowinski, Burnette coined the name “western blot” for his technique. So he developed the method of using electrophoresis to blot the protein onto nitrocellulose paper and after some further work, perfected the technique of blocking non-specific binding sites and visualising the specific radioimmunolabelled antigens using an X-Ray film. Neal Burnette, a post-doc working in the Nowinski group at the Hutchinson Cancer Center in Seattle, started the real fun.īurnette was searching for a way to combine the powers of radio immunoassay and SDS-PAGE electrophoresis so that he could pinpoint specific antigens in a complex protein mixture, such as a cell extract.Īfter some “laughably naive” (his own words – see t his great account by Burnette himself) attempts to visualise the interaction between antibodies and the separated proteins in the gels, he was inspired by Alwine’s nothern blot method (so indirectly by the Southern blot) to make a solid phase replica of the gel. Alwine couldn’t resist the temptation to call his technique the northern blot in an allusion to Southern’s technique, raising a chuckles in labs everywhere. Alwine, a biologist with a sense of humor, developed a technique analogous to the Southern blot, this time for the identification of a specific RNA within a complex RNA sample using a radio-labelled DNA probe. a fractionated genome) and named it after himself – the Southern blot – I’m sure that he had no idea about what he had started. In 1975 when Ed Southern invented his method of using a radiolabeled DNA probe to detect a specific DNA sequence within a DNA sample (e.g. Read on to find out the story of how the Southern, northern and western (etc) blots got their names. It’s a story of discovery, comedy and the triumph of people power over the establishment. This is the story of how one of the most famous and quirky naming conventions in biology came into being. Their marriage is happy as long as they continue to have a good physical relationship. Doctorow writes, “On Sunday afternoon, after dinner, Father and Mother went upstairs and closed the bedroom door”(p 4). The first reference to this is at the very beginning of the novel. After their relationship has been analyzed, one can see that the relationship between Mother and Father is one that seems to be held together purely by sexual desire. ![]() While these scenes definitely imply sexual activity, they are definitely not as intense as the erotic scenes between other characters. The sexual activity that is most relevant to the story line occurs in the relationship between Father and Mother. The sexual activity between Father and Mother is used to define their relationship, sexual activity by Younger Brother is used as a replacement for violence, and the one scene between Houdini and Thaw is used to elucidate the difference between the classes. Some are more obvious and explicit than others, but they all have great purpose. There are three main groups within which the sexual activity takes place: Father and Mother, Younger Brother and Evelyn Nesbit, and Harry Houdini and Harry K. While an obvious reason for these is the titillating effects of them, there are deeper, more relevant reasons for them. Perhaps taking Ragtime into schools and colleges and using it as a teaching tool for race relations will certainly help the next generation understand that it is time to change – the future depends on respect and tolerance for all people, and liberty and justice are meaningless words unless we can live them.Įvelyn Uddin-khan moved to Salisbury in 2018 after living in the New York City area for most of her life. A little bit of tolerance for each other would be a good beginning. It is a sad commentary on we the people that we cannot live in peace, with liberty and justice for all. In fact, after 400 years we have not changed much. It is sad to think that after 120 years we have not changed at all. It was true in 1975, it was true in 1902 and it is true in 2022. It was a poignant reminder that “the more things change, the more they remain the same”.ĭoctorow wrote this story in 1975. Though this can be a result of eye trauma, PVDs are the primary cause of the majority of retinal tears. When the vitreous pulls abnormally (abnormal vitreo-retinal adhesion), it tears the retina as it separates. This usually occurs without any problems but can occasionally cause retinal tears. Initially attached to the retina at birth, this gel gradually detaches from the retina with age, resulting in a posterior vitreous detachment (PVD). ![]() The vitreous fills the retina-lined cavity at the back of the eye. Your risk of permanent vision loss in the affected eye increases the longer retinal detachment remains untreated. The retinal cells are split off from the layer of blood vessels that supplies them with oxygen and nutrients. The term retinal detachment refers to an urgent issue in which the retina, a thin layer of tissue at the back of the eye, moves away from its usual position. ![]() Darkness on the outer edges of your field of vision. We suggest getting in touch with your optometrist right away if you see any of the below changes: When Should You Get Your Eyes Checked?Įye floaters and flashes can be a sign of much more serious eye conditions. People with certain medical conditions such as Sjogren’s syndrome or autoimmune diseases that cause inflammation are at an increased risk for both dry eye disease and floaters. You might even start experiencing them at the same time as you get older. While dry eye disease, as well as eye floaters and flashes, can both be a natural result of the aging of our eyes there is no clear indication one causes the other. Are Dry Eye Disease and Eye Floaters Related? You may see bright flashes as you wake up that fade as the day progresses. Flashes can occur for a variety of reasons but commonly happens when the gel-like vitreous in your eye shrinks naturally with age and begins to pull on your retina.įlashes occur more frequently first thing in the morning or when you are in a dark room. Eye FlashesĮye flashes are bright spots that appear in your line of sight. Floaters move slowly through the vitreous causing them to pass through your sight as they move. These are called eye floaters, and the vitreous in the center of your eye is what causes them.Īs you age, the vitreous within your eye shrinks, resulting in these small shapes. Even when you try to blink them away, they linger around and follow as you shift your gaze. They almost appear to be small bits of dust caught in your vision because they are fuzzy or out of focus. While looking around, you may notice small shapes moving in front of you. However, if you have a lot of floaters and flashes in your eyes, it could be a sign of a serious eye condition called a retinal detachment. Floaters appear as small shapes in your field of vision, whereas flashes can resemble lightning or camera flashes.įloaters are very common and usually do not require treatment. What Are Floaters And Flashes?īoth floaters and flashes are caused by the natural shrinking of the gel-like fluid in your eye (vitreous) as you get older. Women’s Health Study and the Physician’s Health found that dry eye disease incidence rate increases in men and women every five years after the age of 50, with women having a higher prevalence than men. Possible indicators of dry eye disease include:Īging increases your risk factor for dry eye disease. This causes discomfort and irritation in your eyes and can eventually affect your vision. Dry Eye Diseaseĭry eye disease occurs when tears aren’t produced enough to keep your eyes moisturized. Surprisingly, while both issues stem from aging, they are not interrelated. You may have recently been diagnosed with dry eye disease and are wondering if these floaters and flashes are a result of your diagnosis. The next thing you know, the shape is gone and the flash of light has disappeared. You’re going about your day when you see an odd shape make its way across your field of vision, or maybe you caught a flash of light and it’s causing you some concern. ![]() ![]() ![]() North Dakota State College of Science, North Dakota State University. Once the eyeballs are frozen, you can’t stick a skewer through them but what I’d do is just plop them all in the punch to keep it really cold. Then there’s no ice watering down your punch.
